The church was built in Negotin previous to the first rebellion of 1803.

The Church of the Holy Mother was built in 1803, at the time when the Turkish mosque prevailed the worship places of Christian.

However, Pazvan-oglu approved the construction of the church at the request of Metropolitan Kalinik. During the defense of Negotin in the first Serbian Uprising, the church served as a hospital for accommodation of the wounded, and in its port legendary hero Hajduk Veljko and several of his haiduks were buried here.

The famous Serbian composer Stevan Mokranjac also led and sang in the church.
